How thick are these flaps? I have larger tires and not much clearance on wheel well.

These are 5/8" thick. There are off set bracket available that will move the flaps farther away from the tires. You can find those here, https://www.sharptruck.com/truck-hardware-universal-mud-flap-brackets

Will these fit front and rear?

These work best on the front. The full style are best for the rear (GB1223F150). On a 2014 or older F150 use with rear bracket GB755006 with full style flaps.
